DESCRIPTION DE L’APPAREIL
550ºC.
EQUIPMENT DESCRIPTION
The BLOC-DIGEST digestion units are made, mainly,
to do the digestion and determination of Kjeldahl
nitrogen and measurement of C.O.D. Other possible
applications are hydrolysis of samples in determination
of fats, etc.
The digestion units have a heater block, a control unit
and a system to eliminate fumes.
Heater block allocate inside digestion tubes in order
to make an optimal heat transmission and reach a
good temperature distribution among tubes, is
necessary a suitable tubes diameter for the block
that will be used.
Heater block have two safety probes incorporated to
avoid 550ºC overtemperature.

Heater block temperature is controlled by control unit
which is apart of block in order to avoid its damaging
by acid, vapours or splashing. This unit (RAT 2) allows
work until 20 programmes with 4 steps (temperature/
time) in each of them.
In digestion samples to Kjeldahl or hydrolysis stepped
heating allows an optimal foam control because give
the possibility to evaporate humidity of sample before
make digestion at 400ºC.
To remove the fumes produced during digestion you
can use a fume collector together with a water jet pump,
an extraction vent or a scrubber.
Union between fume collector and sample tubes is
not hermetic. This allows that small explosions do not
break glassware. Fume extraction conveys fume to
the waste pipe because of the vacuum that produced
by the water jet pump. So it is essential good operation
of it wich have to do a vacuum level of -0,6 bar.
On C.O.D. technique fumes extraction is not used,
but a special glassware condensate the steam produced
and liquid returns to sample tube.

WORKING OF THE SAFETY THERMOSTAT
If a bad function of the regulation produce an abnormal
increase of the temperature, the electronic safety
thermostat with its second temperature probe, avoid
that temperature reach more than 550ºC. If this
temperature exceed 550ºC for 10 seconds, an acoustic
alarm sounds and the indicator lamp (8) of the control
panel lights up. If temperature low under 550ºC pilot
(8) lights off but alarm condition remain on display.
In case that the failure persists, call up the authorized
technical service.
contenir.
ADVANTAGES OF A STEPPED TEMPERATURE
CONTROL OVER A CONTINUOUS ONE
Continuous process has the problem that when sample
contain humidity this cause foam. This may bring about
losses of sample and damages on metallic block
because of spilts or splashes of acid.
These parameters could differ depending on cain of
sample and the water quantity that it containing.

tenant compte des valeurs orientatives du tableau
de la page 12.
DIGESTION PROCESS:
1.- Connect the control unit and the digestion one.
2.- Place sample tubes into the tube rack.
3.- Introduce into the tubes: samples the catalyst and
the digestion acid.
4.- Move the set at digestion block, using the han-
dles.
5.- Insert the fume exhaust set and the water jet pump
or C.O.D. coolers.
6.- Start up a stored programme in RAT 2 being in
mind page 12 table values .

ENTRETIEN
MAINTENANCE
Before removing the cover disconnect the apparatus
from the mains.
The manipulation by unauthorized people can
cause irreparable damage. Take it to one of the
J.P.SELECTA authorized technical services.
CLEANING:
For cleaning of apparatus use the following products:
7.
11. Laisser refroidir complètement les tubes avant la
distillation.
7.- Push START “7”, if you use a water jet pump for
fumes extraction open the water tap.
8.- Put the lateral plaques on the rack, fixing the screw
in its corresponding hole.
9.- Once the digestion has finished, carefully take out
the tubes with the rack and fume exhaust assem-
bly, leaving them in the rack stand.
10- When the samples have cooled sufficiently, approx.
15 minutes, take out the fume exhaust assembly,
put in its stand and dissolve the samples with dis-
tilled water (normally 50 ml). Mix well, using heat-
resistant gloves for your protection. The cooling of
the tubes can be accelerated by blowing cold air
with a small fan. If the sample is too hot, when you
add the water, the reaction may be too violent and
some of the sample may be lost. On the other
hand, if it is too cold, salts can be precipitated
and this will difficult to dissolve again (solidifications
must be avoided, if this happens, put the tube in
the heater block again for a short time).
11- Leave cool tubes before distilling.

1. Principe:
La méthode consiste à minéraliser
l’échantillon avec l’acide sulfurique
concentré et alcaliniser avec
l’hydroxyde
de
sodium.
L’ammoniaque libéré est éliminé par
distillation et recueilli sur de l’acide
borique. La valoration postérieure à
l’acide chlorhydrique permet le
calcul de la quantité initialement
présente de protéine dans
l’échantillon.
2. Réactifs nécessaires :
• Ácide sulfurique 96% (d=1.84).
• NaOH, solution 35% (p/v).
• Indicateur mixte, spécial pour
titrations d’ammoniaque.
• Catalyseur Kjeldahl
• Acide borique à 4% (p/v).
• HCl 0.25N.
• Eau distillée.
• Pierre ponce en grains.
Note: Il est très important que tous
les réactifs soient totalement
exempts d’azote.
3. Matériel nécessaire :
• Balance de résolution 0.1 mg.
• Unité de digestion (Bloc-Digest).
• Collecteur / Extracteur de fumées.
• Distillateur Pro-Nitro I ou II
• Burette pour titration.
4. Digestion :
chacun
des
tubes
échantillons. (Prendre des
précautions avec la violence de
la réaction).
Laisser refroidir les échantillons à
la température ambiante.
S’il se produit une précipitation,
agiter ou réchauffer légèrement.
5. Distillation
Doser 50 ml d’acide Borique dans
un Erlenmeyer avec quelques
gouttes d’indicateur mixte. Placer
l’Erlenmeyer sur la rallonge du
réfrigérant en prenant la précaution
qu’il reste submergé dans l’acide
Borique.
Une fois mis en place le tube
échantillon et l’Erlenmeyer avec
l’acide Borique, verser 50ml de
NaOH et commencer la distillation.
La distillation doit être prolongée un
temps suffisant pour que se
distillent un minimum de 150 ml,
approximativement de 5 à 10
minutes.
6. Essai en blanc :
Après la distillation d’un échantillon,
Savoir :
P = Poids en g de l’échantillon.
V1= Volume de HCl consommé
dans la titration (ml)
N = Normalité del HCl
V0= Volume de HCl consommé
dans la titration en blanc. (ml)

Introduction
Les méthodes les plus communes
pour
l’estimation
chacune
des
substances organiques présentes
sont les plus laborieuses. Les
méthodes d’analyses globales de
la matière organique contaminant
les usagers actuellement sont :
- La demande chimique d’oxygène
(DCO). Mesure des matières
oxydables par le bichromate de
potassium en milieu acide.
- La demande biologique d’oxygène
DBO). Mesure des matières
organiques biodégradables.
Dans certains pays, la limite légale
de contamination se détermine au
travers de l’expression : ( DBO +
DCO ) / 2
Principe
La demande chimique en oxygène,
D.C.O., peut être considérée
comme une mesure approximative
de la demande théorique
d’oxygène, qui est la quantité
d’oxygène consommée par
l’oxydation totale des constituants
organiques
aux
produits
inorganiques.
La D.C.O. se définit comme la
quantité d’oxygène qui est
équivalente à la quantité de
dichromate consommé par les
matières
dissoutes
et
en
suspension, quand l’échantillon est
traité par le dit oxydant dans des
conditions définies.
Objet et champ d’application
Détermination de la DCO pour les

supérieures à 30 mg/l.
La valeur maximale qui puisse se

déterminer dans un échantillon
sans dissolution est de 700 mg/l.
Cette méthode n’est pas applicable
à des eaux qui après leur
dissolution, contiennent plus de
2000 mg/l d’ions chlorures, comme
l’est l’eau de mer et les eaux
saumâtres.
Description de la méthode
O2/l).
Cette relation est de 1.5 moles
d’oxygène pour chacun de
dichromate..
Réactifs nécessaires

Echantillon
1- Les échantillons pour le
laboratoire doivent être recueillis de
préférence dans des flacon de cris-
tal
ou
polyéthylène.
Les
échantillons sont analysés aussi
rapidement que possible et dans un
délai inférieur à 5 jours. Si les
échantillons sont à conserver avant
de faire l’analyse, on ajoute 10 ml
d’acide sulfurique par litre
d’échantillon et se conserve entre
0ºC et 5ºC. On agite les flacons et
on s’assure que leur contenu fest
bien homogénéisé avant d’en retiret
une partie pour les analyses.
2- Dans le tube à digestion de
D.C.O. 10.0 ml d’échantillon à
analyser. Si la valeur de D.C.O. de
l’échantillon est supérieur à 700
mg/l, on réalise une dilution de
l’échantillon original pour obtenir
une valeurde D.C.O. comprise
entre 350 mg/l et 700 mg/l.
Essai en blanc
3- On réalise un essai en blanc en
même temps que la détermination
, en suivant le même procédé
opérationnel, mais en remplaçant
l’échantillon par10 ml d’eau distillée.
Essai témoin
4- On vérifie la technique et la
pureté des réactifs utilisés pour
l’analyse de 10.0 ml de solution
patron en suivant le même procédé
opératoire que pour l’échantillon.
La demande théorique en oxygène
de cette solution est de 500 mg/l;
le procédé expérimental est
satisfaisant si l’on obtient comme
minimum un 96% de cette valeur.
Détermination
5- On joint 5.0 ml de dichromate
potassium et quelques boules
régulatrices de l’ébullition à
l’échantillon et on homogénéise
soigneusement.
6- On ajoute lentement 15 ml de
sulfate d’argent-acide sulfurique en
agitant avec soin le tube avec un
mouvement
circulaire,
toutes
pertes
de
substances organiques volatiles.
7- On place les tubes préparés
dans la grille porte-tubes sur son
support. Avec l’aide de la grille, on
l’introduit dans le bloc métallique
chauffant.
position D.C.O.
10- On presse "START", à ce
moment commencera à chauffer le
bloc métallique ou sont placés les
tubes, ce sera fait d’une manière
progressive, quand on approche de
la température présélectionnée, le
temps de reflux commencera à
compter.
support.
ferrine. (la couleur doit virer de vert-
bleu à rouge).
Notes:

• NaOH, dilution 35% (p/v).
• mixed of indicator, special for
ammonium titration.
• Kjeldahl Catalyst
• Boric Acid 4% (p/v).
• HCl 0.25N.
• Distilled water
• Pumice stone, granules
Note: It is very important that all
reagent will be free of nitrogen.
3. Material:
• Balance of resolution 0.1 mg.
• Digestion Unit (Bloc-Digest).
• Fume Removal.
• Distillation Unit Pro-Nitro I or II
• Titration Burette.
4. Digestion:
Weigh near 1 g. of sample perfectly
milled and homogenized into a pa-
per exempt of nitrogen and intro-
duced in a digestion tube.
Add to sample tube 10 g. of
Kjeldhal catalyst, 25 ml of sulphu-
ric acid at 96% (d=1.84), and some
granules purnice stone.
Put the digestion tubes with the
sample into the Bloc-digest with
the fume removal working.
Do the digestion at a temperature
between 350º and 420ºC and in a
period of time variable between 1
and 2 hours (see page 12 table).
At the end of the process, the ob-
tained liquid is of green colour or
transparent blue depending on the
used catalyst.
Let cool down the sample at ambi-
ent temperature.
It is important to avoid the precipi-
tation shaking from time to time.
Dosse slowly 50 ml of distillated
water in each tube of sample. (Pay
attention with the violence of the
reaction).
Let cool down the sample at ambi-
ent temperature.
If there is a precipitation, shake or
hot slightly.
5. Distillation.
Dosse 50 ml of Boric acid in an
Erlenmeyer flask and some drops
of mixed of indicator. Put the Er-
lenmeyer below the refrigerant pay-
ing attention to let the output tube
be immersed under the Boric acid.
The distillation has to be extended
the enough time in order to be dis-
tilled a minimum of 150 ml, approxi-
mately between 5 and 10 minutes.
6. Blank:
After the distillation of a sample,
make a blank test, applying the
described method, but using 5 ml
of distillated water instead of sam-
ple.

Introduction
The most common methods in or-
der to estimate the organic con-
tamination are based on the global
determination of oxidizable mate-
ria existing in the water and that
are liable to oxide from the oxygen
of the medium, since the methods
that determine each of organic sub-
stances are complicated. The
methods of global test of organic
materia contaminated used cur-
rently are as follows:
- The chemical request of oxygen
(C.O.D.). Measure of the oxidizable
materia
by
the
potassic
dychromatic in an acid medium.
- The biological request of oxygen
(DBO). Measure of the organic ma-
teria biodegradable.
Some times, the limit of contami-
nation is determinated through the
expression: (DBO+C.O.D.) /2.
Overview:
The chemical request of oxygen,
C.O.D., can be considerated like a
measure approximative of the
theorical request of oxygen, that is
the quantity of oxygen consumed
by the whole oxidation of the
organical constituents to inorganic
products. The C.O.D.. is defined
like the quantity of oxygen that is
equivalent to the dychromat con-
sumed by the dissolved materia
and in suspension, when the sam-
ple is treated by the above oxidant
in the defined conditions.
Target and field of application:
Determination of the C.O.D. for
residual waters with value higher
than 30 mg/l and less than
700mg/l (without dissolution )
This method can be applied to
waters that after being dissolved
contain more than 2000 mg/l of
chlorure ions, like the water of sea
and brackish waters.
Description of the method:
Most of the organical materia is
oxidizable by the effect of a hot
mixture of dychromat and sulfure.
Take the mixture and put back flow
together with the acid solution with
an overrun of the chrome salt
.
After making the digestion, tritate
the overrun with an iron amonium
sulphate (FAS), that allows to know
the oxidizable organical materia,
calculated in terms of oxygen
equivalent (mg O /l).
This relation is of 1.5 mol of oxygen
by each one of dychromat.
Reagents:
Sulphuric acid 4 mol/l: to add to 500
ml. of water, 220 of sulphuric acid .
Let cool down and dilute until 1000
ml.
Sulphuric acid - silver acid: Add 10
g of silver sulphate to 35 ml of
water. Add 965 ml. of sulphuric acid.
Let lie 1 or 2 days in order to be
dissolved. You can help the
dissolution shaking it.

(NH4)2Fe(SO4)2.6H2O 0.12 mol/l
Dissolve 47 g of FAS in water. Add
20 ml. of sulphuric acid (r= 1.84 g/
ml), Let cool down and dissolve with
water until 1000 ml.
This solution has to be contrasted
each day as follows:
- Dilue 10 ml of patron solution of
potasic dichromat until 100 ml. with
sulfuric acid 4 mol/l.
DQO-01 DETERMINATION OF CHEMICAL REQUEST OF OXYGEN (C.O.D.)

standard patron. Dilute 0.4251 g
previously dried at 105ºC, in the
water and fill until 1000 ml with
water.
The solution has a theorical value
of COD of 500 mg/l. This solution
remains stable at least one week if
it is conserved at 4ºC.
Ferroine, indicated solution: 0.7 g
of iron sulphate are dissolved (II)
heptahydratated FeSO4 . 7H2O on
water. Add 1.5 g. of fenantroine -
1,10 monohydratated and it is
shaked until its dissolution. Then
dilute until 100 ml.

Determination:
5. Add 5.0 ml of potasic dichromat
and some glass ballons to regulate
ebullition to the sample and it is
homogenized carefully.
6. Add slowly 15 ml. of silver
suplhate - sulfuric acid, shaking
carefully the tube with a circular
movement, to cool down with cool
water of the tap or an ice bath, in
order to avoid any loss of organical
volatile substances.
7. Put the tubes dully prepared into
the stand tubes holder on their
support. With the help of a stand,
to introduce into the heating bloc.
8. Accoply on each tubes one
refrigerant in order to joint correctly
the polished gaskets.
9. Programm the temperature of
working (170-200ºC) and the time
of lenght (around 2 hours) of the
back flowing into the temperature
regulator and RAT time. Put the
selector of ramp (on the back pa-
nel of the unit RAT ) into the position
C.O.D.
10. Push on «START»and at this
moment the metallic bloc will hot
where the tubes are put. This will
be by a progressive way and when
it reaches the preselectionated
temperature, it will start to count
the time of back flowing.
11. After been finished the time of
back flowing, the heating in the
metallic bloc stops automatically
and you hear an acustic indicator.
At this moment, you should push
on «STOP» in order to finish the
process.
12- To let cool down the tubes for a
while and clean the projections that
have been produced in the inner wall
of the refrigerants into the recipient
of sample with a little volume of
distillated water.
13. To separe the refrigerants from
the tubes, and the tubes go outside
with the help of the stand , that is
put on the support. The sample is
dilued with 75 ml. of distillated water
and let cool down the ambien
temperature.
14. The excess of dichromat is
titred with iron sulphate (II) and of
amonium at the presence of 1 or 2
drops of ferroine . (It has to change
from green-blue colour to red
colour).
NOTES:
When the white is titred, you should
obtain a demand around 10 ml of
FAS.
When you titre the core test you
should obtain a demand of around
4.8ml of FAS.
Material:
C.O.D. equipment composed by:
- Metallic bloc heater.
- Temperature controler
- Tubes for digestion C.O.D.
- Stand for tubes holder.
- Refrigerants for C.O.D.
- Support for refrigerated tubes.
- Burette of precision for titration.
Sampling:
1- The samples for the laboratory
have to be collected preferably into
glass or polyethilene flasks. The
samples are analysed as soon as
possible, never more than 5 days
of their collection. If it is necessary
to conserve the samples before
making the tests, add 10 ml. of
sulfuric acid by 1 liter of sample and
to conserve between 0ºC and 5ºC.
The flasks are shaked and ensure
that
the
content
is
well
homogeneized before taking some
part of the test.
2. To introduce in the digestion tube
of D.C.O. 10,0 ml of the sample to
be analysed. If the value of the
D.C.O. of the sample is higher to
700 mg/l, this is made a dilution of
the original sample in order to obtain
a value of D.C.O. between 350 mg/
l and 700 mg/l.
Blank test:
3. A blank test is done at the same
time that the determination,
following the same operational
procedure, but instead using the
sample, you use 10 ml. of
distillated water.
The theoric demand of oxygen of
this solution is of 500 mg/l; the ex-
perimental procedure is satisfactory
if it is obtained at least 96 % of this
value.
